Abstract

We will establish genetic tools, based around parasexual genetics, which will greatly enhance the experimental tractability of Dictyostelium. We will generate double, triple and more complex mutants, using mitotic recombination for linked genes; set up diploid- based methods for handling disruptions in essential genes; and makes new strains to extend the usefulness of these techniques. We will use these tools to address two hitherto intractable problems. Firstly we make multiple Ras knockouts, using existing single mutants, to test the redundancy and promiscuity of Dictyostelium Ras pathways. Secondly, we will create various gene replacements to allow analysis of the essential actin binding protein Arp2.
