Abstract

The multi-centre proposal builds upon existing research partnerships to create a programme uniquely placed to address advanced problems in the areas of proteomics. In particular, the proposal focuses on the characterisation of intact proteins and protein/protein assemblies, necessitating attention to issues including identifiability (within known-genome species and via cross-species matching), post-translational modifications and protein:protein interactions. New approaches to each stage of protein characterisation will be developed, including procedures for protein separation, analysis using Fourier-transform ion cyclotron resonance (FT-ICR) mass spectrometry, and associated bioinformatic paradigms. The incorporation of FT-ICR method development within the proposed programme of research in proteomics provides a unique opportunity to evaluate and exploit this powerful mass spectrometric approach in the context of post-genome science.
