Abstract
Conjugative plasmids mediating DNA transfer are common in streptomycetes, but little is known about the functions of conjugation proteins such as those of plasmid pIJ101:Tra (for transfer) and SpdAB/KilB (for spread of plasmid through the recipient mycelium). Pilot experiments with SpdA have demonstrated expression in E. coli. This project will go on to study the association of each expressed protein with membranes, plasmid DNA, and other proteins. Alongside biochemistry, genetics will be used to answer questions focussed on spread, including: is Tra needed for spread? What is the DNA specificity of SpdAB/Kil? The outcome will be i) enhanced understanding of natural gene transfer and (ii) a greater ability to manipulate these important cloning vectors.
