Abstract

This grant proposes to use cDNA microarray to identify mRNAs that are translated when cap-dependent translation is compromised. A detailed study will be performed by isolating polysomally associated mRNAs from apoptotic cells as during apoptosis global protein synthesis rates are reduced due to the cleavage of eukaryotic initiation factors by effector caspases. Experiments will be performed to address whether there are changes in the recruitment of specific mRNAs to the polysomes during apoptosis and if this is cell type specific. The signalling pathways that are used in translation initiation during apoptosis will be investigated and finally using in vitro and in vivo reporter assays the mechanisms of translation initiation of specific mRNAs will be determined.
