Abstract
The Escherichia coli MelR protein is a typical member of the AraC family of transcription factors. We propose to investigate the role of the MelR N-terminal melibioise-binding domain in MelR-dependent transcription activation. In order to understand the mechanism of MelR-dependent transcription activation, we will identify the interactions between MelR and the different subunits of RNA polymerase holoenzyme. Finally, in recent work, we have shown that, at the melAB promoter, the cyclic AMP receptor protein is recruited by direct interaction with MelR. We propose to investigate the molecular details of this interaction.
