Abstract

These studies will examine epithelial stem cells in order to a) elucidate physiological cell renewal processes and b) develop methods of stem cell manipulation required for gene therapy and for tissue engineering. Combined techniques of cell culture, retroviral transduction, tissue reassembly and transplantation will be used to determine: a) the size and distribution of stem cell territories in a range of epithelia of differing structural complexity, b) the roles of stromal-epithelial interactions in determining regionally differing stem cell patterns, and c) how stem behaviour is influenced by the expression of genes associated with the control of cell differentiation and death.
