Abstract

Using an inducible KNOTTED1-homebox (KNOX) Arabidopsis line, we have established that gibberellin (GA) will suppress the lobed leaf phenotypes resulting from KNOX misexpression. To further understand this effect, we will study the genetic interactions between Arabidopsis GA mutants and leaf mutants/transgenic lines that misexpress KNOX genes. Similar analyses will be performed using KNOX-loss-of-function mutants. In all cases, GA responsive gene expression will be monitored as well as changes in GA biosynthetic gene expression and GA levels. Finally, we will test whether recreating a hormonal environment of reduced GA and increased cytokinin is sufficient to elicit the lobed leaf phenotype conferred by KNOX overexpression in Arabidopsis. These experiments will provide insight into the way growth regulator and KNOX defined pathways are integrate to control plant development.
