Abstract

Efficient DNA damage responses are essential for the maintenance of genome integrity and cell viability. We have evidence that SUMO-1 modification of proteins is required for some of these responses. We wish to determine the role of SUMO-1 modification using two SUMO- 1 modified targets as test proteins. We will: 1) analyse SUMO-1 modification through the cell cycle and in response to DNA damage and determine whether this is dependent on the DNA integrity checkpoint proteins; 2) identify and mutate the SUMO-1 modification sites in our test proteins and analyse the phenotypes of resulting mutants; and 3) investigate stability and localisation of the modified test proteins and ask whether modification affects protein-protein interactions.
