Abstract

The overall aim is to understand how FNR-dependent promoters that carry a DNA site for FNR near position -41 are organised, to understand how these promoters differ from CRP-dependent promoters with a similar architecture, and to understand the mechanisms by which FNR-dependent transcription activation is down-regulated by upstream-binding factors. Working with an FNR- dependent promoter carrying a DNA site for FNR near position -41, we propose to: 1) identify the determinants of alpha CTD that make contact with FNR during transcription activation; 2) determine the location of the two alpha CTDs and the organisation of transcriptionally-competent complexes; 3) investigate the consequences of binding a second FNR molecule upstream; 4) determine the mechanism by which upstream-bound FNR inhibits FNR-dependent transcription activation; and 5) investigate the consequences of upstream binding of CRP or FIS.
