Abstract
The work outlined here is designed to determine the molecular mechanism of a genetic switch and to elucidate and understand the structural and mechanistic interplay between three proteins and two small molecules. My laboratory is in an ideal position to address these issues. Our in vitro reconstruction of galactose regulation represents a major advance in the understanding of the switching mechanism. We now have at hand all the tools required to dissect this system and unravel the protein-protein and small molecule interactions that play a vital role in controlling transcription. Understanding the precise details of a specific genetic switch will lead to a better comprehension of how and why aberrant gene expression occurs.
