Abstract

We have developed a novel assay based on surface plasmon resonance for monitoring molecular recognition events and dynamic processes during in vitro transcription on chromatin templates. We propose to exploit this assay to a) probe the roles of a hormonally regulated bZIP transcription factor in the initiation of transcripts at the abscisic acid (ABA) regulated Em promoter; b) probe similar events controlled by a family of GATA factors at the light regulated rbcS-1A promoter, and c) probe the role of 'aberrant' RNAs in gene silencing.
