Abstract

The cap-binding complex eIF4F plays a pivotal role in eukaryotic translation initiation. We will: 1. use genetics, SPR analysis and X-ray crystallography to characterise interactions within the eIF4F core complex (eIF4E:eIF4G) and between eIF4F and Poly(A)-binding protein (Pab1) and the regulatory eIF4E-binding proteins (p20/4E-BP); 2. determine the molecular mechanisms underlying the heterotrophic cooperativity modulating eIF4E-cap binding and the regulation of eIF4F via p20/4E-BP; 3. examine the global and mRNA- specific modulation of translation in response to changes in eIF4F activity caused by eIF4F mutations, nutrient-limitation and stress; and 4. test for cyclical eIF4F exchange during initiation using reconstruction methods.
