Abstract
The basiodiomycete dikaryon represents an extended stage between mating cell fusion and nuclear fission. Two signalling pathways ensure that each cell maintains a nucleus from each mate, an intracellular one triggered by the A mating type genes and an intracellular one triggered by the B mating type genes. Using a combination of DNA-binding studies and REMI mutagenesis, we propose to elucidate key steps in the A pathway and ask whether any of these are shared by the B pathway. We will exploit our knowledge of two key transcriptional factors which are implicated in activating the A pathway, a homeodomain protein heterodimer encoded by the A mating type genes and an HMG domain protein encoded by pcc1, to specifically select mutations in genes that are likely targets of the A protein heterodimer and to distinguish these from genes encoding downstream components of the pathway.
