Abstract

We have identified a temperature sensitive mutation (mlc1-1) in a gene coding for a putative myosin light chain, MLC1. Preliminary phenotypic observations suggest that the protein plays a role in regulating actin/myosin cytoskeletal function. It is proposed to undertake a detailed cytological study of the terminal phenotype to establish the precise role of Mlc1p within the cell. This will include localisation of functional, monoclonal epitope tagged Mlc1p. We will characterise the proteins with which Mlc1p interacts using both genetic and biochemical approaches. Thereafter the possibility that the protein is regulated by calcium-dependent phosphorylation will be tested by genetic and biochemical experiments.
