Abstract

Yeast and mammalian cells regulate gene expression at the level of translational initiation via proteins that bind the cap-binding protein eIF4E. We have recently established a new system for studying these eIF4E-binding proteins (4E-BPs) in S. cerevisiae which can be used to examine their mechanism of action in considerable detail. We will: 1) Characterise the relationship between binding-domain structure and the eIF4E-binding affinity and specificity of the respective yeast and human 4E- BPs (p20 and 4E-BP1) using techniques of mutagenesis surface plasmon resonance and X-ray crystallography; 2) Determine how the binding of p20/4E-BP1 to eIF4E in yeast modulates translation in vivo and characterise the mechanism regulating this interaction; 3) Use the yeast-based regulatory system to screen for mammalian genes that encode kinases capable of modulating 4E-BP function.
