Abstract
Our long-term aim is to test the primary actions of the Pax-6 gene in regulating neural development. Currently existing mutants do not allow us to distinguish primary and secondary actions of Pax-6. We shall use the Cre recombinase-loxP system to generate new tissue and stage-specific null-mutants. We shall generate mice carrying loxP sites in the Pax-6 gene (thereby sensitising Pax-6 to the mutagenic action of Cre recombinase). We shall cross them with mice carrying the Cre recombinase gene under the control of a telencephalon-specific or inducible promoter. We shall examine the phenotypes of the resulting mutants and examine the effects of inducing knock-outs at different ages.
