Abstract

The U5 snRNA is required for alignment of exons for the second step of pre-mRNA splicing. The mechanisms by which U5 recognises and becomes positioned with the exons at splice sites are unclear. This project will establish whether the U2 and U6 snRNAs, the proposed catalytic factors in splicing, interact with U5 to direct it to the exons. U2 and U6 mutants that block the second step of splicing will be analysed to determine if they influence exon alignment of U5 in vitro. Direct interactions between U5 and these snRNAs will be mapped at high resolution by site specific crosslinking in vitro and investigated by genetic studies in vivo. Such analysis will further our understanding of the critical cellular process of pre-mRNA splicing.
