Abstract

MelR is a transcription activator belonging to the AraC family of bacterial transcription factors. We propose to use a combination of molecular genetics and biochemistry to investigate four fundamental questions concerning the function of MelR (and other members of the family): (1) how does MelR interact with DNA? (2) what is the role of MelR binding at different target sites? (3) how does MelR interact with RNA polymerase to activate the melAB promoter? (4) does MelR interact with other transcription factors?
