Abstract

The key objective of this proposal is to achieve the molecular cloning and characterization of the GEMINI80 gene which is required for correct division asymmetry and cell fate determination at pollen mitosis I (PMI). The specific aims are (1) to use map based cloning and targeted transposon-tagging to isolate the GEMINI80 gene; (2) to monitor precisely the developmental events leading to altered division symmetry in gem80, including analysis of nuclear migration, spindle orientation and cytokinesis; and (3) to use GUS and GFP marker gene technologies to monitor how altered division symmetry affects cell fate in gem80; and (4) to determine allelism between gem80 and five related gemini mutants.
