Abstract

Transcription activation at Class I promoters dependent on the Escherichia coli cyclic AMP receptor protein (CRP) is dependent upon a single direct interaction between a surface- exposed activating region in CRP and a contact site in the C-terminal domain of the RNA polymerase alpha subunit (alphaCTD). We propose to identify the contact site for this activating region in alphaCTD, to quantify CRP:alphaCTD interactions and to investigate the consequences of this interaction. Finally, we plan to investigate the organisation of alphaCTD subunits at promoters carrying the DNA site for CRP at different locations upstream of the RNAP-binding elements.
