Abstract

We aim to begin the systematic isolation and functional analysis of all members of an important plant transcription factor gene family - MADS-box genes. Plant MADS-box genes encode many structurally related proteins which control development. The work forms two overlapping parts: isolation and function search. Gene isolation will be by homology methods (mixed probe low stringency screening, PCR), computer searches (other species, ESTs) and non-homology based screens for MADS protein interactions. Function search will be by PCR selection of loss-of-function mutants from large populations. Genetic interactions will be determined by crossing the obtained mutants with each other and with existing mutants.
