Abstract

The eukaryotic cap-binding complex eukaryotic initiation factor eIF4F mediates the interaction of ribosomes with mRNA, thus playing a key role in translational initiation. It is also a focal point of regulation of gene expression. We will use a combination of genetic and biochemical methods to study the function and regulation of this complex in both budding and fission yeast. The experiments will include: (i) mutational and competition analysis of eIF4F component interactions in Saccharomyces cerevisiae; (ii) in vitro analysis of mammalian-type regulatory circuits in S. pombe; (iii) in vitro biochemical studies of eIF4F interactions using cell-free translation systems and recombinant eIFs and eIF domains.
