Abstract

We aim to characterise a set of cue (CAB underexpressor) mutants of Arabidopsis, previously isolated by the applicant in the laboratory of J Chory (Salk Inst.), to identify one primarily affected in either light or plastid signalling regulating nuclear photogene expression. To this end, in each cue mutant, we will: a) Examine the morphology and a marker protein in etioplasts. b) Analyse photogene expression under diagnostic conditions. c) Attempt to rescue the phenotypes of cues with a gun1 mutation, which partly uncouples nuclear photogene expression from plastid functional status. d) Use a set of available CAB3 promoter fragment:luciferase reporters, to identify promoter regions through which each cue mutation acts, and compare them with regions responsible for plastid regulation. This project will define the target for a subsequent one, centred on cloning one CUE gene.
