Abstract
The objective of this proposal is to generate improved hybrid-constructs capable of targeting high-level protein production to mammary cells. This will be achieved by incorporating RNA sequence elements from hepatitis B virus S gene, herpes simplex virus thymidine kinase gene and the bovine preprolactin gene into constructs encoding either human alpha-1- antitrypsin or factor IX. Mammary specificity will be directed by ovine beta-lactoglobulin sequences. The expression potential of genomic- and cDNA-based modified constructs will be evaluated by comparison to the parental constructs. A comparison of their ability to enhance protein production by stable transfection of cultured cell lines in vitro and in transgenic mice in vivo will enable a rationale design of future expression vectors which will be broadly applicable in all aspects of gene transfer technology.
