Abstract

The CCAAT motif is found in the promoters of a quarter of eukaryotic genes and several binding activities for it are known. A CCAAT box transcription factor (CBTF) undetectable in nuclei or on chromatin until GATA-2 transcription begins. Evidence strongly suggests that CBTF is novel. We will analyse the DNA-binding, subunit interaction and transcription activation domains of CBTF. We will use deletions, point mutants and chimaeras of CBTF made in E. coli and embryos. Thus we will understand how CBTF's functions are affected by developmental regulation, the mechanism of its action and the relationship of CBTF to other CCAAT factors.
