Abstract

Comparative and mutational analysis of the Thr-Gly repeat of the PERIOD (PER) protein in Drosophila melanogaster has revealed that it plays a major role in the temperature compensation of the circadian clock, and that this may be mediated by interactions with other regions of PER. Molecular and immunological methods will be used to examine the circadian dynamics of PER and TIMELESS (TIM-the PER partner), and their association in these mutants, both in vivo, and in vitro. In addition we will examine these putative interactions using both the yeast two-hybrid system and in vitro associations of the relevant PER peptides. The results will illuminate the molecular basis of circadian temperature compensation.
