Abstract

Preliminary work is described which shows that yeast whi2 mutants fail to enter Go, while whi2p overexpression leads to filamentous growth. In whi2 mutants G1 cyclin synthesis is deregulated during starvation, accounting for the continued division observed. Two mutants, gin3 and gin6 have been isolated which are synthetically lethal with whi2 and probably act in the PCK1/MAP kinase pathway. The GIN3 and GIN6 genes will be cloned and characterised. Possible genetic interactions of WHI2 with members of the PCK1/MAP kinase pathway CLN3, BCK2, GRR1, ELM1-3 and PHD 1 will be investigated. Biochemical approaches based on epitope-tagged Whi2p will be used to examine its intracellular location, possible interaction with the G1 cyclins or the p34 kinase, DNA binding and its effects on CLB synthesis or activity. Finally, the function of a possible actin-binding motif will be assessed.
