Abstract

If transposition could be confined to meiosis, every germinal transposition recovered in progeny would be unique. To refine plant transposon tagging strategies, the Arabidopsis homologue (AtDMC1) of the yeast DMC1 meiosis-specific gene has been isolated. The AtDMC1 gene promoter confers a meiosis-specific pattern of GUS reporter gene expression. This promoter has been fused to the Ac and Spm transposase genes, and to the Tnt1 retrotransposon. Transgenic Arabidopsis plants carrying these constructs will be tested for meiosis-specific transposition events.
