Abstract

There is conflicting evidence regarding the chromosomal distribution of HMG14/17 and HMG1/2. We aim to resolve these discrepancies using experience gained in mapping acetylated core histones. The methodology is to crosslink intact cells with formaldehyde (to avoid disturbance of native structures), isolate DNA/protein adducts on CsCl gradients, immunoprecipitate chromatin fragments using affinity-purified polyclonal antibodies, reverse the crosslinks and then probe the DNA with unique genomic sequences from the gene/locus under study.
