Abstract

Me1R is an E coli transcription activator. Our objectives are: (1) to find the mechanisms by which active Me1R stimulates transcription initiation. To do this we will isolate mutations resulting in constitutively-active Me1R, purify the mutant protein and study DNA binding and transcription activation in vitro. (2) to obtain structural data on Me1R by overexpressing and purifying the C-terminal DNA-binding domain and initiating an NMR study of the structure. (3) to locate segments of Me1R involved in the triggering of activity by melibiose.
