Abstract

Pluripotent haemopoietic stem cell proliferation and development is regulated by a family of cytokines. One such cytokine Granulocyte Macrophage Colony Factor (GM-CSF) binds to a cell surface receptor to elicit both proliferative and developmental responses in progenitor cells. To identify the receptor domains associated with proliferation and development respectively, mutated GM-CSF receptors will be transferred into a multipotent stem cell line using retroviral gene delivery system. The cellular signalling events associated with proliferative and/or developmental responses will then be identified using these ectopically expressed receptor mutants.
