Abstract
Our objective is to discover how bacterial c-type cytochromes are assembled from haem and apoprotein precursors and targeted to the E. coli periplasm. Mutants and cloned genes already isolated in this laboratory will first be characterised. A mutant deleted for six recently- identified genes will be constructed and complementary DNA will be cloned from a phage library. Three probes for protein targeting or haem attachment will be used to identify genes for a haem lyase, how haem or cytochromes are transported across the membrane and the site of haem attachment.
