Abstract
We have identified both the regulatory (acu-1) and the structural gene (acs-1) for the enzyme acetyl-CoA synthetase in the mushroom Coprinus cinereus. Our observation that a mutation in acu-1 can cause temperature sensitive activity of the acs-1 enzyme indicates that there is a direct interaction between the two proteins. We aim to 1. Isolate and sequence the acu-1 gene. 2. Study its own regulation by Northern analysis and promoter analysis. 3. Use in vitro modification and expression to study its regulation of acs-1 and one of the key enzymes of the glyoxylate pathway (acu-7). 4. Ask whether acs-1 has an independent regulatory role in regulating enzymes of the acetate utilisation pathway.
