Abstract
The clones of a microlibrary derived from microdissected B- chromosomes will be characterised with respect to copy number, B- specificity, sequence homology and chromosomal location. Parts of the B-chromosome will be microdissected in order to enrich for clones containing desirable sequences. Gene expression and modulation of transcriptional activity of B-chromosomes will be explored by differential screening of cDNA libraries. A detailed and thorough understanding of the molecular organisation and genetic activity of the B is seen as a preliminary to its evaluation and development as a plant artificial chromosome for the transformation of rye and related species.
