Abstract

Initiation of translation of some eukaryotic viral RNAs eg EMCV, FMDV occurs by direct entry at a site internal within the RNA, the so-called internal ribosome entry segment or IRES. By exploiting the power of yeast molecular genetics the role of both cis- and trans-acting factors, important for viral IRES function, will be defined. In addition, endogenous cellular yeast mRNAs which are translated via an IRES will be identified and characterised and a comparative analysis of internal initiation in yeast and higher eukaryotes undertaken. This work will provide important new insights into this unusual mode of translation and, in a wider context, to a range of biological problems including virus tissue tropism and disease resistance.
