Abstract

Large numbers of human genes are being sequenced and identified. Assignment of function is lagging far behind. We propose to use the model organism Saccharomyces cerevisiae to attack this problem by constructing a panel of yeast strains, in each of which, an essential gene is placed under the control of a down-regulatable promoter. The panel will be transformed with human cDNA libraries retrofitted into a yeast expression vector. We will down-regulate expression of the endogenous yeast targets and look for rescue (or partial rescue) by complementation. Examples of anti-complementation will also be assayed. The procedures will allow the categorisation of the function of human genes. (Joint with grant FGT11381).
