Abstract

An enhancer trap based on Ac/Ds transposons and designed to generate patterns of expression of the synthetic transcription factor LhG4 has been constructed. LhG4 expression is detected by GUS and GFP genes under control of LhG4-dependent promoters at an unlinked site. Any gene can then be mis-expressed in the same pattern as LhG4. Arabidopsis plants carrying the constructs have been. We propose to test the enhancer trap by generating 5000 lines containing transposed elements. Published data suggest that 48 percent of these will drive patterns of LhG4 expression, and this will be tested by analysing GUS expression in 3000 lines, and GFP expression in a subset of these. Sequencing DNA adjacent to 300 insertions will locate them within the genome and identify the genes underlying some of the expression patterns. This will provide transposon-based mis- expression technology in Arabidopsis that given the wide host range of Ac/Ds will be readily transferable to crop plants. (Joint with grant FGT11322).
