Abstract

We request support for a 200kV electron microscope and associated cryotransfer and specimen preparation equipment. This will create a facility for high resolution electron microscopy of biological molecules within the biological structure group in the Krebs Institute at Sheffield University. Major research programmes which will use the new facilities will involve structure determination of membrane proteins from 2D crystals including components of bacterial photosynthetic apparatus, eukaryotic channel proteins, bacterial antibiotic transporters and viral membrane fusion proteins. Structure determination at low resolution of non-antibiotic transporters and viral membrane fusion proteins. Structure determination at low resolution of non- crystalline protein complexes will include analysis of the ruv complex involved in homologous recombination.
