Abstract

We aim to develop new mathematical tools for analysing protein and gene expression time series in order to investigate the structure and behaviour of gene regulatory networks. This will be done in the context of a specific biological system: the regulatory network centred on p53 that controls programmed cell death in response to irradiation induced DNA damage. We will measure time series of the expression of 12,500 human genes using DNA microarray technology and of the levels and activities of key regulatory proteins. We will then construct and analyse a sequence of increasingly sophisticated models of the regulatory network which will be used to identify the key components of the network, their roles, and interactions, and the effects of damage or mutations to them on cell apoptosis or survival. This will be done in an iterative fashion with ongoing experiments.
