Abstract
Early differentiation of embryonic stem cells will be analysed by microarray expression profiling. Amplification of the target RNA for use from very small tissue samples will be validated and will be applied to isolated tissues from microdissected embryos and from cells sorted from in vitro differentiation. This will allow resolution of the complexity of profiles from the mixtures of cell types found both in vitro and in vivo. Specific differentiation and its manipulation particularly along the neuronal pathway will be studied. An experimental genetic analysis will be undertaken using pre-existing resources of ES cells with gene knockouts and, most importantly to close the experimental loop, using locus-specific knockdown.
