Abstract
Hyperbranched polymers including dendrimers and dendronised linear polymers offer certain advantages as nano-sized drug carriers. They are monodisperse and display unique 3D architecture. We will investigate systematically the effect of polymer structure on endocytosis, intracellular trafficking and transcytosis with a view to optimising the design of dendrimer-based drug and protein delivery systems. A library of dendrimer and dendronised polymer probes will be synthesised and their mechanisms of endocytosis and subsequent trafficking quantitated using subcellular fractionation and molecular cell biology techniques. Structures will be optimised to promote and drug delivery by each pathway.
