Abstract

Developing generic reagents for high throughput studies of protein- protein interactions is the main goal of this research. Though attractive, cross-linking of protein complexes requires extensive optimisation as well as prior knowledge of at least one member of a multiple protein assembly. Molecular design and chemical synthesis will be used to create molecules that overcome these barriers. Prototypes will be tested against known protein complexes in vitro, assessed by gel electrophoresis and mass spectrometry and improved. Application of these agents to cells followed by affinity purification and general global analysis is the ultimate objective.
