Abstract

In this project, a synergistic combination of in vitro experiments and mathematical modelling will be used to optimise the efficacy of a novel, macrophage-based gene delivery system. This exploits the tendency of macrophages to migrate into hypoxic regions of tumours to deliver hypoxia-regulated transgenes to such areas. We will determine strategies that optimise macrophage migration into hypoxic areas, gene delivery, and therapeutic impact, by identifying the potential role of: (i) specific macrophage attractants released by tumour cells; (ii) changes in the levels of receptors for these factors expressed by macrophages; (iii) pre-therapy levels of macrophages in hypoxic tumour areas; (iv) bystander effects.
