Abstract
The TIBS tissue engineering programme aims to establish a scientific basis (platform technologies) for the design and operation of advanced mechanical tissue bioreactors, growing neotissues as mechanical units. The exemplar target tissue is a ligament-bone construct. The 4 project consortium (part of a wider London collaboration: ALTEC) comprises UCL (lead), QMC and IC. Existing, novel mechano-bioreactors, based on pioneering work in cytomechanical regulation, will be used to solve the twin problems of nutrient perfusion and collagen deposition and organisation. These will establish basic mechanisms and the role of (i) COMBINATION cues of mechanical loading and growth factors; (ii) background factors - existing matrix and gravity; (iii) novel (hard and soft) composite cell support biomaterials; (iv) use of rational cell and matrix control cues for the new design platforms to test neo-tissue growth (a) long term (b) as integrated 2-phase units (ie scientifically rational advanced bioreactors).
