Abstract

We have isolated a cell from the surface of articular cartilage that has the properties of a progenitor:stem cell. We wish to further characterise this cell in terms of its requirements for growth factor:morphogenes for it to progress through its differentiative repertoire, particular attention being focussed on the expression of cell fate selection genes Notch 1 and its ligands Delta and Jagged. We will determine whether the progenitor-like properties are lost with age and if this is the case whether they can be rescued by transfection techniques. Finally, we will gain insights into the role of the EDA variant of fibronectin that is selectively synthesised at the articular surface in regard to possible mitogenic activity.
