Abstract
In this proposed project, continuous sampling techniques using micro membrane probes and analysis protocols to monitor cell viability and functionality across three dimensional constructs will be developed based on microdialysis and ultrafiltration. It is a minimally invasive and non-destructive method. The simple and widely studied constructs of using poly L-Lactic Acid (PLA) scaffold seeded with bovine chondrocytes will be used as a model to test the developed methods. The developed monitoring techniques would enable us to measure (a) the local concentrations of glucose and oxygen (to monitor nutrient supply), lactate (to monitor cell metabolic activity) across the constructs, (b) the collagen synthesis and matrix turnover, and (c) the extracellular matrix accumulation within the construct during culture.
