Abstract

Numerous cytochrome P450 (CYP) gene sequences have been uncovered by genome sequencing projects which confronts biologists with the task of identifying the function of the gene products. We shall exploit our newly developed substrate-trapping technology to determine the roles that the CYP superfamily play in Streptomyces coelicolor A3(2) by: (i) heterologous expression in Escherichia coli and purification of each CYP; (ii) determination of CYP production within S. coelicolor through Western blot analysis; (iii) identification of each CYP substrate through the unique technology of substrate-trapping. This approach will be generic for the functional genomic analysis of the cytochromes P450 superfamilies across all the Kingdoms of Life and through identification of their natural substrates, and will allow the efficient exploitation of such enzymes in biotechnology.
