Abstract
The aims of this project are: 1) To develop a novel method for determining the average mutation rates in directed evolution (DE) experiments based on sloppy PCR; 2) Thereby to calibrate and optimise the mutation rate in three DE systems; 3) To establish the optimal size of DNA fragments in directed recombination in these systems; and 4) Finally, therein systematically to determine the independence or otherwise of evolution at separate sites on a protein.
