Abstract

An on-line probe for precise measurement of oxygen flux will be combined with heat flux as a powerful monitor of aerobic and anaerobic processes in animal cells growing in bioreactor culture. The value of the metabolic probes will be demonstrated by their detection of the decisive mitochondrial events in environmentally-initiated apoptosis in hybridomas. This will be contrasted with the protection afforded to cells by the overexpression of the bcl-2 protein in cells. Metabolic changes will be correlated with events well-documented by flow cytometry. The ability of exogenous agents to suppress chemically-induced apoptosis in continuous culture will be metabolically monitored to reveal clues to both mechanism and possible restorative strategies. The probes will reveal the metabolic consequences of overcoming bcl-2 protein protection with chemotherapeutic drugs.
