Abstract
The DNA mismatch binding protein MutS will be fused to the nuclease domain of the type IIs restriction enzyme FokI, generating a hybrid protein which recognises and cleaves mismatch-containing DNA fragments. We will optimise the activity of this fusion protein by altering the linker between the two domains and by using truncated version of MutS. This chimeric protein will have many uses in DNA diagnostics and molecular biology and we will examine its use for removing DNA mismatches generated during PCR and for detecting single base mutations.
